Olive leaf extract impairs mitochondria by pro-oxidant activity in MDA-MB-231 and OVCAR-3 cancer cells

Breast and ovarian cancers are the leading fifth reason for tumor death among females, respectively. Recently, many studies demonstrated antiproliferative activities of natural aliments in cancer. In this study, we investigated antitumor potential Olive Leaf Extract (OLE) triple-negative breast cancer cells. A HPLC/DAD analysis on OLE has been performed to assess total polyphenolics other secondary metabolites content. HCEpiC, MDA-MB-231, OVCAR-3 cell lines were used. MTS, Cytofluorimetric, Western Blot analyze viability, proliferation, apoptosis, oxidative stress. Fluorimetric IncuCyte® analyses carried out evaluate apoptosis mitochondrial function. We confirmed that OLE, containing a quantity oleuropein 87 % extract, shows anti-proliferative pro-apoptotic activity MDA-MB-231 For first time, our results indicate inhibits viability inducing cycle arrest, it also increases apoptotic up-regulating protein level cleaved-PARP caspase 9. Moreover, data show treatment causes significant decrease functionality, paralleled by reduction membrane potential. Interestingly, increased intracellular reactive oxygen species (ROS) together with decreased ROS scavenging enzymes, confirming stress both models. Our demonstrate generation represented primary mechanism activity, as pretreatment antioxidant N-acetylcysteine prevented OLE-induced arrest apoptosis.
